What supplements should I take for MTHFR gene variations?

The core of an MTHFR protocol is methylcobalamin, methylfolate, and SAMe. Methylcobalamin and methylfolate are the final bioactive forms of B12 and folate - they require no conversion by the MTHFR enzyme.

What does SAMe actually do?

SAMe (S-adenosylmethionine) is the molecule that donates methyl groups to over 200 biochemical reactions, including neurotransmitter synthesis, DNA methylation, phospholipid production, and detoxification. B12 and methylfolate regenerate SAMe through the methionine cycle.

Can methylated B vitamins cause anxiety?

Some people with COMT gene variations process methyl groups slowly. For them, high-dose methylated B vitamins and SAMe can temporarily raise catecholamine levels - dopamine and norepinephrine - which may feel like anxiety or restlessness.

SAMe should not be combined with SSRIs, SNRIs, MAOIs, tramadol, or other serotonergic medications due to risk of serotonin syndrome. SAMe may trigger mania in people with bipolar disorder.
